It sounds possible on paper, but in real life it can become very rushed. You may spend more time inside the vehicle than actually enjoying the places.

Airport / Negombo → Sigiriya → Kandy → Nuwara Eliya or Ella → Yala or Udawalawe → South Coast → Airport

This gives a good mix of culture, nature, wildlife, mountains and beach. But even this route should be adjusted based on your flight time, hotel style, children, age, travel month and how much relaxation you want.

Another important point is beach selection. Some travellers try to add both south coast and east coast in the same short trip. But these areas are far apart. For a 10-day Sri Lanka tour, it is usually better to choose one beach region instead of trying to cover both sides. This makes the holiday smoother and more enjoyable.
